Introduction
L'activité scientifique du groupe sarcomes de l'Institut Bergonié bénéficie d'une reconnaissance internationale, avec des publications dans tous les domaines du diagnostic et de la prise en charge thérapeutique et une forte présence dans les congrès internationaux tels que l'ASCO et le congrès Nord-Américain de pathologie (USCAP).
En 2011, 2 études ouvertes promues par l'Institut étaient financées par un PHRC et une autre par un contrat NCI-INCa. L'Institut Bergonié maintient une collaboration importante avec les groupes académiques français, européens et américains (GSF-GETO, EORTC, SARC notamment), des partenariats privilégiés avec des institutions françaises (CRLCC, équipes INSERM) notamment dans le cadre du cancéropôle GSO, ou étrangère tels les groupes de Louvain ou du MSKCC avec des projets déjà finis et d'autres en cours. Le groupe dispose aussi de partenariats industriels et ainsi, d'un accès à la plupart des nouveaux médicaments testés dans les sarcomes.
L'intégration des activités de recherche biologiques et cliniques permet et va permettre d'évaluer en clinique la pertinence de concepts novateurs conçues dans la section sarcome de l'U 916, tels que l'inhibition de MDM2 dans les liposarcomes ou anti AURORA-K. de par ses activités scientifiques et par son rôle de coordination nationale dans les réseaux mis en place par l'INCa RREPS et NETSARC, l'Institut Bergonié poursuit sa mission de structuration de la recherche sur les sarcomes en France : mise en place et gestion des bases de données, intégration de l'étude moléculaire dans la prise en charge des patients notamment (projet soumis au PHRC 2012).

Essai CHONDROG
Étude multicentrique de phase II évaluant l'efficacité de GDC-0449 chez les patients atteints d'un chondrosarcome localement avancé et/ ou métastatique.

Objectif principal : Évaluation de l'activité anti-tumorale de GDC-0449 en termes de bénéfice clinique à 6 mois, la réponse étant définie selon les critères d'évaluation de la réponse tumorale (critères RECIST 2009, v1.1)

Étude multicentrique de phases IIb/III comparant l'efficacité de la trabectédine administrée en perfusion de 3 heures ou de 24 heures et de la doxorubicine chez des patients atteints de sarcome des tissus mous avancé ou métastatique non traité.

Objectif principal : Évaluer si la trabectédine, administrée en chimiothérapie de 1ère intention dans le sarcome des tissus mous avancé ou métastatique, prolonge la survie sans progression par rapport à la doxorubicine.

Essai TOMOREP
Évaluation de l'association radiothérapie en tomothérapie hélicoïdale (54 Gy) suivie de chirurgie dans les liposarcomes rétropéritonéaux.

Objectif principal : Évaluation de l'efficacité de l'association radiochirurgicale avec irradiation par tomothérapie hélicoïdale pour une dose de radiothérapie de 54 Gy chez des patients atteints d'un liposarcome rétropéritonéal opérable. L'efficacité sera évaluée en termes de survie sans récidive locale à 3 ans.
